코로나19 팬데믹 발생 이후, 백신, 체외진단 그리고 치료제에 대한 R&D 투자가 활발하게 이루어지고 시장이 확장됨에 따라 전세계적으로 바이오의약품 시장은 급격한 성장세를 보이고 있다. 지난해, 국내 바이오산업의 생산과 수출 또한 역대 최대 증가율을 기록하였다.

 

바이오의약품이란?

 바이오의약품은 사람이나 다른 생물체에서 유래된 것을 원료 또는 재료로 하여 제조한 의약품으로 생물학적제제, 유전자재조합의약품, 세포배양의약품, 첨단바이오의약품 및 기타 식품의약품안전처장이 인정하는 제제를 포함한다. 바이오의약품은 합성의약품보다 분자량이 크고 구조가 복잡하여 제조과정에 어려움이 있으나 부작용이 적고 표적에 직접적으로 작용하여 우수한 약효를 나타낸다. 코로나19 사태가 발생한 이후, 합성의약품이 풀지 못한 난제를 바이오의약품이 해결하면서 더욱 주목을 받고 있다. 더불어 효과적인 치료제가 아직 개발되지 않아 미충족 수요가 많은 희귀난치성질환의 치료제 개발에 대한 기대가 높아지면서 바이오의약품의 중요성이 대두되고 있다.

 

코로나19 팬데믹 이후의 바이오의약품 시장

 코로나19로 인해 최초의 mRNA 백신이 역사상 가장 빠른 속도로 개발되었다. mRNA를 투여하면 체내에서 스스로 바이러스의 스파이크 단백질을 생성한 후, 면역시스템이 작동하여 이에 대한 항체를 생산하도록 유도하는 방식으로 작용한다. mRNA 백신은 바이러스를 직접 주입하는 백신들에 비해 독성이 낮아 안전성이 높고, 바이러스의 유전자 정보를 통해 빠르게 대량생산할 수 있어 개발기간이 짧다는 장점이 있다. mRNA 기술은 오래전부터 연구되어 왔기 때문에 기초 연구의 기반이 잡혀 있었고, 이에 더하여 코로나19 상황이 백신 개발에 있어서 결정적인 부스터 역할을 한 것이다.

 

 전세계에 타격을 입힌 코로나19로 인해 백신, 진단키트, 그리고 치료제에 대한 수요가 증가하면서 바이오의약품 시장의 입지는 점차 넓어지고 있다. 생명공학정책연구센터의 발표에 따르면 코로나19 팬데믹 이후, 바이오의약품 시장규모는 2021 5837억 달러에서 2027 9113억 달러까지 성장할 것으로 전망되고 있다. 이는 코로나19 이전, 2027년의 시장 규모가 7034억 달러로 예상되었던 것과 비교하였을 때 약 2000억 달러 이상이 상향 조정된 수치이다. 또한, 글로벌바이오시장의 연평균 성장률이 코로나19 이전에는 6.2%였으나 팬데믹 상황이 지속되면서 7.7%까지 증가하였다. 코로나19 상황이라는 위기를 기회로 삼아 바이오의약품 시장은 더욱 성장하고 있으며, 코로나19 백신과 치료제 개발에만 머무르지 않고 다양한 희귀난치성질환 치료를 위한 바이오신약 개발의 가속화에 기여할 것으로 예상된다.

 

바이오의약품 개발과 AI 기술과의 융합

 코로나19 팬데믹 이후, 바이오의약품 시장이 급성장할 수 있었던 주요 요인 중 하나로 ‘AI 기술과 융합을 꼽을 수 있다. 코로나19 백신과 치료제의 후보 물질 발굴에도 AI 기술이 적극 활용되고 있다. AI 기술은 유전체 데이터를 바탕으로 바이오마커 혹은 약물이 작용하는 타겟을 찾아내는 것뿐만 아니라 질병을 진단하고 예후를 예측하는 단계에서도 활용된다. 다양한 상황을 컴퓨터 시뮬레이션으로 처리하여 결과를 예측할 수 있는 AI 기술을 통해 후보 물질 발굴 단계의 시간을 단축할 수 있다는 것이 큰 강점으로 작용한다.

 

 구글의 자회사인 딥마인드는 아미노산 서열 정보만으로 단백질의 3차원 구조를 예측할 수 있는 알파폴드2”를 개발하여 2020년 초 코로나19 원인 바이러스인 SARS-CoV-2와 미지의 영역에 있던 ORF3a 구조를 예측했다고 밝혔다. 또한, 모더나는 표적에 대한 치료용 항체를 식별하는 AI 기술의 도입을 위해 올해 9AI 기반 항체 발굴 플랫폼을 보유하고 있는 캐나다 ‘AbCellera’와 계약을 맺어 연구를 진행하고 있다. 이처럼 코로나19AI 신약의 시대를 앞당기고 있으며, 여러 기업들이 뛰어들어 바이오의약품 분야에서 의미 있는 성과를 내고 있다.

 

 최근 국내 기업들도 인공지능 기반 개발업체와 손을 잡고 바이오의약품 신약후보물질 발굴에 박차를 가하고 있다. mRNA 개발에 핵심적인 5’-캡핑 및 지질나노입자(LNP) 전달 플랫폼을 제공하는 에스티팜과 유전자 서열분석을 통한 신항원(Neo-antigen) 예측법으로 mRNA 항암백신 후보물질을 도출하는 AI 기술을 보유한 테라젠바이오가 신항원 mRNA 항암백신의 연구개발 및 CDMO(위탁개발생산) 협력에 나섰다. 암환자로부터 추출한 암세포 유전자의 변이와 특성을 AI 기술로 분석하여 최적화된 신항원을 예측한 후, 이를 암호화하는 mRNA를 암환자에게 투여해 암세포에만 특이적으로 면역반응을 일으켜 암을 치료하는 것이다. 이외에도 안지오랩’, ‘노바셀테크놀로지등 수많은 국내 기업에서 혁신적인 바이오신약 개발의 가속화를 위해 AI 기술과의 융합을 추진하고 있다.

 

국내 바이오의약품 산업의 전망

 코로나19로 인해 전세계적으로 수요가 늘어남에 따라 바이오의약품에 대한 투자가 확대되고 있으며, 첨단기술과의 융합으로 개발 속도가 빨라져 바이오의약품 신약 파이프라인은 지속적으로 늘어날 것으로 보인다. 글로벌 바이오의약품 산업이 발전함에 따라 국내 기업 또한 경쟁력을 확보해 나가야 한다. 과학기술정보통신부의 2022년 바이오 원천기술개발 투자 계획에 따르면 총 투자금액 5532억원 중 신약 분야에 837억원을 투자할 예정이라고 한다. 이처럼 바이오산업은 국내에서 유망한 산업으로 각광받고 있으며, 정부의 지원이 끊임없이 확대되고 있다. 해외 기업과의 협력 및 공동연구개발과 AI 기술을 포함한 다양한 분야와의 융합까지 효과적으로 이루어진다면 국내 기업도 바이오시밀러 개발에서 더 나아가 포스트 코로나 시대의 혁신적인 바이오의약품 개발을 이끌 수 있을 것이라 기대한다.

 

Rapidly growing biopharmaceuticals after the COVID-19 outbreak

 

 After the COVID-19 outbreak, the biopharmaceutical market has shown rapid growth worldwide as R&D investments in vaccines, in vitro diagnosis, and treatments have been actively made and the market has expanded. Last year, the domestic bioindustry production and exports also recorded the highest growth rate ever.

 

What are biopharmaceuticals?

 Biopharmaceuticals are medicines manufactured using raw materials or materials derived from humans or other organisms and include biological agents, gene recombination drugs, cell culture drugs, advanced biopharmaceuticals, and other drugs approved by the Minister of Food and Drug Safety. Biopharmaceuticals have a larger molecular weight and a more complex structure than synthetic drugs, making them difficult to manufacture. However, they have fewer side effects and directly act on targets to show excellent medicinal effects. Since the COVID-19 incident, biopharmaceuticals have been attracting more attention as they have solved the challenges that synthetic drugs have not solved. Moreover, the importance of biopharmaceuticals is emerging as expectations for the development of treatments are rising for rare and incurable diseases with high unmet demand.

 

The biopharmaceutical market after the COVID-19 pandemic

 Due to COVID-19, the first mRNA vaccine was developed at the fastest rate in history. The mRNA vaccine acts in such a way that when mRNA is administered, it generates a spike protein of the virus itself in the body. Then, it induces the immune system to produce antibodies against it. It is safer than vaccines that directly inject viruses due to less toxicity. Also, it has the advantage of a short development period as it can be mass-produced quickly through genetic information of viruses. mRNA technology has long been studied and laid the foundation for basic research, and in addition, the COVID-19 situation has served as a decisive booster in vaccine development.

 

 The biopharmaceutical market is gradually expanding as demand for vaccines, diagnostic kits, and treatments increases due to COVID-19, which spread across the globe. According to an announcement by the Biotechnology Policy Research Center, after the COVID-19 outbreak, the biopharmaceutical market is expected to grow from $583.7 billion in 2021 to $911.3 billion in 2027. This is an increase of more than $200 billion compared to the market size before COVID-19 which was expected to be $703.4 billion in 2027. Also, the average annual growth rate of the global bio market was 6.2% before COVID-19 but increased to 7.7% as the pandemic continued. Taking advantage of the crisis of the COVID-19 incident as an opportunity, the biopharmaceutical market is growing further, which is expected to contribute to accelerating the development of new biopharmaceutical drugs for the treatment of various rare and incurable diseases.

 

Convergence of development of new biopharmaceuticals with AI technology

 One of the major factors that allowed the biopharmaceutical market to grow rapidly after the COVID-19 pandemic is "fusion with AI technology." AI technology is being actively used to discover candidate materials for COVID-19 vaccines and treatments. AI technology is used not only to find biomarkers or targets for drugs based on genetic data, but also to diagnose diseases and predict prognosis. One of the strengths of AI technology, which can predict results by handling various situations with computer simulations, is that it can shorten the time of the candidate material discovery stage.

 

 Google's subsidiary company ‘DeepMind’ developed "AlphaFold 2", which can predict the three-dimensional structure of proteins only with amino acid sequences. It predicted the structure of SARS-CoV-2, a virus that causes COVID-19, and the ORF3a structure at the beginning of 2020. In September, ‘Moderna’ signed a contract with Canada ‘AbCellera’, which has an AI-based antibody discovery platform, to introduce AI technology that identifies therapeutic antibodies to targets. As such, COVID-19 has advanced the era of new AI drugs, and various companies are jumping in and making meaningful results in the biopharmaceutical field.

 

 Recently, Korean companies have also joined hands with artificial intelligence-based developers to accelerate the discovery of new drug candidates. ‘ST pharm’ and ‘Theragen Bio’ have decided to cooperate in joint R&D and CDMO. ‘ST pharm’ provides a 5'-capping and lipid nanoparticle (LNP) delivery platform essential for mRNA development, and ‘Theragen Bio’ has AI technology to derive candidate substances for mRNA anticancer vaccines through genetic sequencing. First, the mutations and characteristics of cancer cell genes extracted from cancer patients are analyzed with AI technology to predict optimized neo-antigens. Afterward, the mRNA encoding them is administered to cancer patients to treat cancer by causing an immune response that responds specifically to cancer cells. Furthermore, numerous domestic companies such as ‘AngioLab’ and ‘Novacell Technology’ are pushing for convergence with AI technology to accelerate innovative biopharmaceutical development.

 

Prospects of the domestic biopharmaceutical industry

 As the global demand increases, investment in biopharmaceuticals is expanding and the speed of the development is increasing due to COVID-19. Therefore, biopharmaceutical pipelines are expected to increase consistently. As the global biopharmaceutical industry grows, domestic companies should also enhance competitiveness. According to the Ministry of Science and ICT's 2022 bio-source technology development investment plan, 83.7 billion won will be invested in the new drug sector out of the total investment of 553.2 billion won. As such, the bio-industry is in the spotlight as a promising industry in Korea, and government support is constantly expanding. If cooperation with foreign companies, joint R&D, and convergence with various fields including AI technology is effectively carried out, it is expected that domestic companies will be able to lead the development of innovative biopharmaceuticals in the post-COVID-19 era.
